=== Amber codon suppression === The possibility of reassigning codons was realized by Normanly et al. in 1990, when a viable mutant strain of E. coli read through the UAG ("amber") stop codon. This was possible thanks to the rarity of this codon and the fact that release factor 1 alone makes the amber codon terminate translation. Later, in the Schultz lab, the tRNATyr/tyrosyl-tRNA synthetase (TyrRS) from Methanococcus jannaschii, an archaebacterium, was used to introduce a tyrosine instead of STOP, the default value of the amber codon. This was possible because of the differences between the endogenous bacterial syntheses and the orthologous archaeal synthase, which do not recognize each other. Subsequently, the group evolved the orthologonal tRNA/synthase pair to utilize the non-standard amino acid O-methyltyrosine. This was followed by the larger naphthylalanine and the photocrosslinking benzoylphenylalanine, which proved the potential utility of the system. The amber codon is the least used codon in Escherichia coli, but hijacking it results in a substantial loss of fitness. One study, in fact, found that there were at least 83 peptides majorly affected by the readthrough. Additionally, the labelling was incomplete. As a consequence, several strains have been made to reduce the fitness cost, including the removal of all amber codons from the genome. In most E. coli K-12 strains (viz. Escherichia coli (molecular biology) for strain pedigrees) there are 314 UAG stop codons. Consequently, a gargantuan amount of work has gone into the replacement of these.

== Enzyme structure == Enteropeptidase is a type II transmembrane serine protease (TTSP) localized to the brush border of the duodenal and jejunal mucosa and synthesized as a zymogen, proenteropeptidase, which requires activation by duodenase or trypsin. TTSPs are synthesized as single chain zymogens with N-terminal propeptide sequences of different lengths. These enzymes are activated by cleavage at the carboxyl side of lysine or arginine residues present in a highly conserved activation motif. Once activated, TTSPs are predicted to remain membrane-bound through a conserved disulfide bond linking the pro- and catalytic domains. In the case of cattle enteropeptidase the primary translation product comprises 1035 residues with an expected mass of 114.9 kDa. The detected apparent mass of about 160 kDa is consistent with the specified carbohydrate content of 30 - 40%, with equal amounts of neutral and amino sugars. The activation cleavage site after Lys800 splits the heavy and light chains of mature cattle enteropeptidase. There are 17 potential N-linked glycosylation sites in the heavy chain and three in the light chain; most of these are conserved in other species. The heavy chain has a hydrophobic section near the N-terminus that supports the transmembrane anchor. The heavy chain influences the specificity of enteropeptidase. Native enteropeptidase is resistant to soybean trypsin inhibitor. However, the isolated light chain is subtle whether prepared by limited reduction of the natural protein or by mutagenesis and expression in COS cells.
Lactobacillus delbrueckii subsp. lactis is a subspecies of Lactobacillus delbrueckii that is generally used to measure the amount of cobalamin in food. Its growth rate is proportional to the amount of cobalamin in the growth medium. However, lactis has been demonstrated to have the option to utilize pseudocobalamin, which is inactive for humans, as well as "alkali-resistant factors" (deoxyribosides and deoxynucleotides), leading to an overestimation of the amount of cobalamine in food. As such, new methods using HPTLC or LC-MS/MS have also been developed.

Concurrent use or unregulated dosages of piperacillin results in increasing levels of piperacillin within the body, prolonging neuromuscular transmission blockages created by non-depolarizing muscle relaxants, and disruptions in urine tests for glucose. Some compounds that may interfere with the bactericidal activity of piperacillin include chloramphenicol, macrolides, and sulfonamides. Following two studies conducted in 1986 and 2006, piperacillin was found to inhibit the removal of methotrexate in animal kidneys. Furthermore, in the presence of piperacillin-tazobactam, the decay time for methotrexate triples in comparison to the normal half-life, leaving the patient exposed to cytotoxic effects produced by the chemical agent. While penicillin antibiotics generally work synergistically with aminoglycosides by enhancing their penetration of bacterial membranes, they can also work adversely by inactivating them. A reformulation of ethylenediaminetetraacetic acid and piperacillin-tazobactam has produced results showing an increase in their affinity with amikacin and gentamicin in vitro, enabling the process of simultaneous Y-site infusion to occur.
